Tramadol hydrochloride, a synthetic opioid, acts via a multiple mechanism of action. Tramadol can potentially change the behavioral phenomena. The present study was undertaken to investigate the effect of tramadol on learning memory activity using novel Object of accessing short term memory. Twenty Five (25) male mice weighing between 25g - 35g were employed in the present investigation. Novel object apparatus served as the esteroceptive behavioural models. Twenty five male albino mice weighing around 25–35?g were employed in the present investigation. The animals were randomly divided into five groups. group 1 received distilled water (10 ml/kg, p.o.) and served as control, group 2 received tramadol (25 mg/ml, p.o), group 3 received tramadol (50 mg/ml, p.o.), group 2 received tramadol (75 mg/ml, p.o.), while 5 was received tramadol (100 mg/kg, p.o) for 14 days consecutively. The result obtained from the tramadol treated mice shows a dose dependent decreased in d2 and R indices when compared to the control group. These results indicated that the tramadol-treated group spent less time for exploration of the novel object than the familiar object. Notably, the severity of damage was notice at high dose of tramadol (100 mg/kg). It was further observed that the administration of tramadol as an atypical opioid analgesic in a low dose or high doses impairs the memory in mice. The observed effects can be justified according to the unique properties of tramadol in the opioid family. It was concluded that administration of tramadol may disturb learning and memory and so should not be recommended for use without doctor prescription.
